Haploidentical HSCT in Hemoglobinopathies in the Gene Therapy Era: Current Strategies, Broader Access

Allogeneic hematopoietic stem cell transplantation (HSCT) has represented for long the only established cure for thalassemia and sickle cell disease. With the recent approval of gene therapy products, the therapeutic algorithm for hemoglobinopathies is being reshaped, raising questions about patient selection, donor availability, and long-term outcomes.

The introductory talk will review current indications for allogeneic HSCT, summarize outcomes from matched and alternative donor transplants, and outline how the field is adapting to a more crowded therapeutic landscape.

The two main talks will then focus on the haploidentical transplantation and on the two major platforms: αβ T-cell depleted haploidentical transplant and PTCy-based haploidentical transplant — as strategies to overcome the persistent barrier of donor availability.
