Background:
Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by recurrent colonic inflammation and substantial morbidity. Effective management requires drug delivery that targets the colon while limiting premature release in the upper gastrointestinal tract. Vardenafil, a phosphodiesterase-5 inhibitor with reported anti-inflammatory activity, is a promising repurposed candidate for UC; however, its poor aqueous solubility and limited bioavailability may restrict therapeutic usefulness. Nanocarrier-based systems can improve drug incorporation, protect the drug during gastrointestinal transit, and enhance colonic localization.
Aim:
This study aimed to develop, optimize, and dual-coat vardenafil-loaded PLGA polymersomes with chitosan and Eudragit S100 to provide sequential mucoadhesive, pH-responsive, and microbiota-responsive delivery to the colon.
Methods:
Vardenafil-loaded PLGA polymersomes were prepared by emulsion solvent evaporation. A response surface design optimized the polymersomes in terms of the particle size and the entrapment efficiency. The optimized polymersomes were further coated with chitosan and Eudragit S100, then characterized for particle size, polydispersity index, zeta potential, and entrapment efficiency. The optimized dual-coated formulation was evaluated for in-vitro release, mucoadhesion, and pharmacokinetic performance.
Results:
The optimized system formed dual-coated nanoparticles with a particle size of 206.17 ± 12.6 nm, PDI of 0.29 ± 0.01, zeta potential of +49.65 ± 9.54 mV, and entrapment efficiency of 70.43% ± 7.41. Drug release was limited in simulated gastric and intestinal fluids, with 19.8% ± 1.8 released within 5 h, followed by sustained release reaching 97.7% ± 2.8 after 24 h in simulated colonic fluid. Mucin interaction increased particle size 5.32-fold, indicating mucoadhesion. Compared with standard vardenafil, the nanoparticle formulation increased relative bioavailability 3.52-fold and prolonged residence time 7.19-fold.
Conclusion:
Chitosan/Eudragit S100 dual-coated PLGA polymersomes provide a promising platform for colon-targeted vardenafil delivery. Their sequential coating combines mucoadhesive, pH-responsive, and microbiota-responsive properties, improving controlled release, and residence time, and supporting further investigation for UC therapy.
