Quantitative proteomics research has demonstrated to be essential to understand how biological systems work at a molecular level. However, the bioinformatic analysis and processing of this enormous amount of data for meaningful biological and functional interpretation remains to be challenging. Here, we will discuss various current standardized methods and strategies for bioinformatic analysis, and we will highlight some available software programs, computational tools, and meta-analysis resources for proteomic data analysis. |

Show Abstract |
|||||||||||||||||||||||||||||||||||||
Cancer immunotherapies targeting the PD-L1/PD-1 axis are revolutionizing cancer treatment and transforming the practice of medical oncology. Despite the recent successes, most patients are refractory and present intrinsic or acquired resistance. This is one of the most significant challenges in oncology. However, the mechanisms leading to resistance are still poorly understood. Here, we discuss some of the major molecular mechanisms of resistance to PD-L1/PD-1 blockade. We also argue whether tumor intrinsic or extrinsic factors constitute main determinants of response and resistance. Primary and acquired resistances are important barriers in terms of benefit to the patient. Among others, tumor-intrinsic factors include poor tumor antigenicity, DNA repair mutational load, alterations in the regulation of oncogenic pathways, defects in IFN signal transduction and PD-L1 expression. On the other hand, tumor-extrinsic factors include T cell exhaustion, expression of additional immune checkpoint molecules, differentiation and expansion of immunosuppressive cell populations, and release of immunosuppressive cytokines and metabolites. The relative contribution of tumor cell intrinsic and extrinsic factors to primary, adaptive, and acquired resistance is yet unclear. Our group recently found that NSCLC patients that failed to respond to PD-1/PD-L1 inhibitors had systemic CD4 dysfunctionality, characterized by increased PD-1/LAG-3 co-expression in T cells. These results highlighted dysfunctional CD4 T cells as main contributors to treatment failure. A deeper understanding of the basic mechanisms underlying resistance will provide insight for further development of better therapeutic strategies by overcoming treatment failure. The development of new approaches to improve CD4 responses before immunotherapy could be a solution to overcome resistance (Zuazo et al, EMBO Mol Med 2019; Zuazo et al, EMBO Mol Med 2020; Chocarro de Erauso L et al, Front. Pharmacol. 2020; Zuazo et al, Font Immunol 2020; Bocanegra et al, Int J Mol Sci 2020; Hernández et al, Int J Mol Sci 2020; Arasanz et al, Cancers 2020; Chocarro et al, Int J Mol Sci 2021). |

sciforum-060454 | Role of the next-generation immune checkpoint LAG-3 in response and resistance to cancer immunotherapy | , , , , , , , , , , , | N/A |
Show Abstract |
|||||||||||||||||||||||||||||||||||||
Lymphocyte activation gene 3 (LAG-3) is a cell surface inhibitory receptor with multiple biological activities over T cell activation and effector functions. LAG-3 plays a regulatory role in immunity and emerged some time ago as an inhibitory immune checkpoint molecule comparable to PD-1 and CTLA-4 and a potential target for enhancing anti-cancer immune responses. LAG-3 is the third inhibitory receptor to be exploited in human anti-cancer immunotherapies, and considered a next-generation target in cancer immunotherapy, right next to PD-1 and CTLA-4. Unlike PD-1 and CTLA-4, the exact mechanisms of action of LAG-3 remain poorly understood. Indeed, PD-1/LAG-3 co-expression is a marker for exhausted T lymphocytes infiltrating tumors. This constitutes a high-risk signature, but also a clinical factor associated to overall survival. The lack of understanding of LAG-3 functions is partly caused by the presence of non-conventional signaling motifs in its intracellular domain, different from others present in classical immune checkpoints. Here we summarize the current understanding on LAG-3 and its role in response and resistance to cancer therapy, from its mechanisms of action to clinical applications (Chocarro de Erauso L et al, Front. Pharmacol. 2020; Zuazo et al, Font Immunol 2020; Bocanegra et al, Int J Mol Sci 2020; Hernández et al, Int J Mol Sci 2020; Arasanz et al, Cancers 2020; Chocarro et al, Int J Mol Sci 2021). |

|||||||||||||||||||||||||||||||||||||
Lymphocyte activation gene 3 (LAG-3) is a cell surface inhibitory receptor with multiple biological activities over T cell activation and effector functions. LAG-3 plays a regulatory role in immunity and emerged some time ago as an inhibitory immune checkpoint molecule. A systematic research was performed using PubMed and ClinicalTrial.gov databases. Up-to-date published articles meeting the inclusion criteria were investigated. LAG-3 expression has been linked to increased pathology in certain inflammatory disorders, such as HDL Hypercholesterolemia and Inflammatory Bowel Disease. LAG-3 protein expression levels have been significantly associated via transcriptomic studies with high HDL cholesterol (HDL-C) and HALP (HDL-C ≥ 60 mg/dL). Indeed, LAG-3 deficiency altered lipid raft formations and cell phosphosignaling, processes leading to an enhanced proinflammatory state, and increased production of inflammatory cytokines such as TNFα (Golden et al, Insight 2016; Rodriguez et al, Curr. Atheroscler. Rep. 2021; Chocarro et al, Int J Mol Sci 2021). LAG-3 has been reported to be a modulator of T cell regulation in inflammatory responses in the intestine (Do et al, Mucosal. Immunol. 2016; Chocarro et al, Int J Mol Sci 2021). In addition, LAG-3+-regulatory T cells are required to suppress the inflammatory activities of CX3CR1+ macrophages to maintain tissue homeostasis during lymphoid cell-driven colitis (Bauché et al, Immunity 2018; Chocarro et al, Int J Mol Sci 2021). Moreover, LAG-3+ cells have been shown to be increased in the inflamed mucosa and correlate with endoscopic severity and disease phenotype in ulcerative colitis. Here, we will discuss the impaired control of cell-mediated immunity associated with high accumulation of LAG-3 in inflammatory disorders (Chocarro et al, Int J Mol Sci 2021). Interestingly, in vitro blockade of PD-1/LAG-3 interactions enhances cytokine production in response to cancer and infections, and it is demonstrating promising results in several clinical trials for the treatment of various cancers, suggesting it could have a similar effect in inflammatory disorders (Chocarro et al, IOTECH 2022). A deeper understanding on the basic mechanisms underlying LAG-3 intracellular signaling will provide insight for further development of novel strategies for autoimmune and inflammatory disorders (Chocarro et al, Int J Mol Sci 2021). |

LAG-3 protein expression has been shown to correlate with increased coronary heart disease and increased myocardial infarction. LAG-3 accumulates in cardiac allografts undergoing rejection episodes to fully vascularized, heterotopic, allogeneic heart transplantation (Haudebourg et al, Transplant 2007; Chocarro et al, Int J Mol Sci 2021). LAG-3 deficiency has also been associated in clinical studies with increased risk of coronary artery disease due to Tr1 dysfunction (Zhu et al, Hum. Here, we will discuss the impaired control of cell-mediated immunity associated with high accumulation of LAG-3 in cardiovascular diseases (Chocarro et al, Int J Mol Sci 2021). 05. NICE.XSM-08: North-Ibero-America Congress on Exp. & Simul. Methods, Valencia, Spain-Miami, USA, 2022
Welcome Message
We are glad to inviting you to NICE.XSM-08: North-Ibero-America Congress on EXperimental & Simulation Methods, Valencia, Bilbao, Spain - Miami, USA, 2022. Development of new experimental and computational methods are sometimes separated areas but more often runs hand-by-hand. In fact, Computer-assisted experimental techniques are of the major importance nowadays in Chemistry, Medicine, Economics, Engineering, Industry, Epidemiology, etc. For instance HPLC, NMR, PCR, X-Ray devices used in chemistry, medicine, food industry, transport, are all of them computer-assisted devices. The development of these devices implies a previous research in both the validation of the experimental technique (physics, chemistry, molecular biology) and the development of a user-friendly computer-assisted device (Mathematics, Data Analysis, Artificial Intelligence, Computer Engineering). Both research may run independently or totally coordinated from the very beginning.
Topics. Topics range from research on development and application of pure experimental methods (physics, chemistry, materials, nanotechnology, biotechnology, medicine, etc.) to pure computational simulation methods and/or further integration of both classes of methods on computer-aided experimental methods. The fields of application include, but are not limited to, Biomolecular Sciences, Biomedical Engineering, Materials, Environmental, Trade, Learning, Education, and Social sciences in general (see scope section). This is an inter-university transatlantic joint congress co-chaired by USA and Europe institutions. In first instance, the founder chairs are professors of the Institute of Molecular Science (ICMol), University of Valencia (UV), and the Catholic University of Valencia (UCV), Valencia, Spain. From the America's side the workshop is co-hosted by professors of Department of Mathematics Miami Dade College (MDC) and Department of Chemistry and Biomolecular Engineering, Tulane University (TULANE), New Orleans, USA. All in all, the workshop is associated to the MOL2NET International Conference Series on Multidisciplinary Sciences. MOL2NET (From Molecules to Networks) series is running this year MOL2NET-2021, International Conference on Multidisciplinary Sciences, ISSN: 2624-5078, MDPI SciForum, Basel, Switzerland, 2021. Consequently, the present congress is co-chaired and promoted as well by researchers of the University of The Basque Country (UPV/EHU) and IKERBASQUE, Basque Foundation for Science, Bilbao, Basque Country.
Experimental methods. Made special emphasis on experimental methods and computer-assisted experimental methods in bio-molecular sciences and biomedical sciences. Examples of experimental methods of interest are those used on Biophysics, Molecular Biology, Medicinal Chemistry, Biomaterials, Biotechnology, Biomedical Engineering, etc. such as Microscopy, Spectroscopy (NMR, IR, Raman, X-Ray) for Biomolecules Characterization, Image recording methods (fNMRi), EEGG and ECG, etc.
Computational methods. This term includes, but is not limited to, Computational Biophysics, Computational Biology, Quantum Chemistry, Bioinformatics, Systems Biology, Complex Networks, Discrete Mathematics, Mathematical Analysis, Complex Networks Sciences, Statistics, Data Analysis, Artificial Intelligence (AI), Machine Learning, Deep Learning, etc. Last, not the least, the congress deals with all the Legal, Regulatory, and Bioethics issues emerging from the use of both experimental and modelling and simulation methods in the previous areas. This include, Drug, Gene, Protein, Vaccine, Biomarkers, and Biomaterials patentability, Personalized Medicine and Personal Data Protection, Cybersecurity, Cryptocurrency Mining, Data Protection in Personal Image Processing, Software and Models Copyrights, etc.
NICE.XSM Target Audience. This congress focus on both under-graduated, MSc, and PhD students, as well as Professors, Engineers, Researchers, and Industry partners. The topics of interest cover both Experimental and Computational Modelling and Simulation Methods along with Data-driven Decision Making in Industry and Engineering.

MOL2NET Transversal Associated Workshops. DEMAI'22 is a MOL2NET Associated Workshops are special workshops published transversally in more than one MOL2NET congresses according to the specific topic of each paper. They are invited directly by the chairman, and/or proposed by others member of the committee, or collaborators who become chairmans of the workshop proposed by themselves. Consequently, these workshops may appear announced in more than one congress page. The authors of the communications submitted to this class of workshop have to select one of congress of the conference to submit their papers according to their criteria. The conference chairman reserves the right of reallocating the paper to other congress according to the topic. Please, see at follow the special workshop(s) associated to this congress.
DEMAI'22 topics: In the new age of the Fourth Industrial Revolution or Industry 4.0 (4IR), digital transformation and exponential change are well-known trends in the business sector. These trends are driven by the vast amount and diversity of data being generated around the world every second. Data has become so important to business that it is being called "the new gold standard" and coined the popular concept of "Data-Driven Decision Making" meaning that decision makers and stakeholders need to consider the data in business decision processes. The two most important actors in a decision process are the decision makers and the stakeholders. Where stakeholders provide valuable insights, judgments, recommendations that support decision makers in making decisions, but to be successful they must incorporate the insights provided by data into their preferences, which must be aligned with the mission and strategic vision of the business. We welcome contributions related to the following fields: Decision Sciences, Data Science/Analytics, Business Intelligence/Analytics, Expert Systems, Artificial Intelligence/Machine Learning (AI/ML), Deep Learning, Statistical Learning, Operations Research, Mathematical modelling, Behavioral and Organizational aspects of the decision making process. Data Engineering, Decision Engineering.
DEMAI'22 objectives: The objective of this event is to provide a space for practitioners, scientists, analysts and professionals who work or are related to decision making in companies and organizations to share experiences, ideas, scientific work and applications related to decisión support from a broader perspective including data driven decision making, decision maker focused, considering tangible (quantitative) and intangible (qualitative) factors that influence decision making to develop effective decision support strategies (Decision Making Engineering).

08. REVIEWWWERS.PUB-08, Reviewers & Pre-Post Publication Workshop, Bilbao-Miami, USA, 2022.
REVIEWWWERS = Review + World Wide Web (WWW) Pre-Post Publication (PUB) Workshop
Welcome to REVIEWWWERS.PUB-08, Reviewers & Pre-Post Publication Workshop, Bilbao-Miami, USA, 2022. This the special workshop has two phases. Phase 1: focus on review of early view abstracts. Phase 2: focus on pre-prints and post-publication review, per se.
Phase 1: Early View Abstracts Review phase: This first phase starts with the beginning of the conference and endures all the year.It focus on the processing of abstracts submitted to the conference using SCIFORUM platform. We publish all pre-accepted abstracts until full presentation files are uploaded, approved, and published in the proper congress. We also keep in this buffer section, permanently published online, all abstracts of papers submitted to the conference and accepted preliminarily but without full communications files uploaded. Please, scroll down to until the end of this Abstracts and Workshops session and click on toggle list link to see/hide the list of accepted abstracts without full communications uploaded.
Authors liability. Remember that our conference publish only pre-print papers and post-print/post-publication summary content. It means that, this conference is a way for authors to communicate their earlier results (pre-print) and/or discuss already published results from their/others groups (post-print discussion). Consequently, in all cases, it is responsibility of authors, to ensure veracity and originality of the content. Authors are recommended to performing a similarity checking, ensure veracity of contents, and carry out a proper citation of previous works. In this sense, we strongly recommend the authors to use online text-similarity checking services, if possible. These tools may help authors to avoid any form of plagiarism or copyright violation.
Committee liability. The committee is not responsible of the previous aspects (similarity, veracity, proper citation) in this publishing modality. Consequently, the opinions expressed on the communications are not necessarily coincident to the opinions of the members of the committee. The authors may be requested to modify the communication (re-write their texts) in the case that high similarity is detected by a third part and reported to the committee. The committee may decide to take-down from public web (unpublishing) a communication if sufficient evidence of violation of ethic veracity or originality principles is reported according to committee criteria. In these cases, the manuscript could be put on standby or withdrawn temporarily until the authors re-submit the proper version. In the post-print/post-publication discussion communications the authors are also allowed to submit short reviews, comments, letters, critical assays, opinions, or discussions of papers already published. The scientific committee of the early view publication workshop is composed by selected advisory experts worldwide and some section chairpersons of different congresses who have participated in the pre-screening of abstracts.
Phase 2: Post-Publication Review Phase. This the online post-publication review phase of the MOL2NET'22, Conference on Molecular, Biomedical, & Computational Sciences and Engineering, ISSN: 2624-5078, MDPI SciForum, Basel, Switzerland, 2022. This year we attracted >470 participants including committee and authors, and also >170 communications and abstracts submitted to >10 USA-Europe Inter-University Congresses and Workshops. Now, we are closing early view abstracts review phase and also publication phase of the conference. Consequently, we are now in the post-publication review phase of the conference. This last phase, usually runs on the first month of the new year. In this phase we cele . This is a workshop focused on the post-publication discussion/review of all the papers published in the last edition of the conference. The workshop also focus on post-publication abstracts indexed in our conference with full versions papers already published as part of special issues associated to the conference and published by MDPI journals. In this workshop we invite all colleagues, authors, committee, and/or validated conference followers to participate by posting comments to communications, questions to authors, answers/suggestions to reviewers and followers, public collaborations proposals, etc. You can become a certified post-publication reviewer acting over the web. The participation in the workshop is totally free of cost. We wish you a fruitful reviewwwing experience!!!
Reviewwwers role. The Sciforum platform, host of this conference, allows you to perform post-publication review of papers. MOL2NET conference opens all years a post-publication stage. On this stage each year we prompt reviewers worldwide to perform post-publication reviews/comments about of papers published the year before and interact online with the authors of all these communications. By getting involved you can become a certified reviewer of conference communications acting over the web. One this modality of the workshop we invite all members of committee, authors, and validated social network followers, to become post-publication reviewers of all papers published the year before in our conference. They can make public questions to authors, suggest new collaborations, etc.
Inter-congress cross-commenting. Cross-commenting of authors of one workshop to other workshop are specially welcome. People invited are professors, researchers, PhD/MSc students, and science writers with validated LinkedIn, ResearchGate, ORCID, or Publons profiles. We aware our reviewers to take into consideration that most papers published are preliminary pre-print versions. Consequently, the objective is not to perform a detailed review of the papers. The objective is to call the attention of authors over interesting points, do recommendations to improve the paper, propose collaborations, etc. Comments will be moderated by our staff. We also offer webinars and free of cost certified reviewers training. Please, follow these steps to patriciate.
Instructions to Post-publication reviewwwers. Step (1), Register/Login to Sciforum platform. Step (2), Go to [MOL2NET'22 Papers List], with all papers published last year. Step (3), Scroll down papers list and click on title to go to the paper you selected. Step (4), Scroll down click on Commenting button post your comment and click submit.
